A reverse transcriptase - polymerase chain reaction based assay for Borrelia species detection in ticks was developed. The method was based on amplification of 552 nucleotide bases long sequence of 16S rRNA, targeted by Borrelia specific primers. In the present study, total RNA extracted from Ixodes ricinus ticks was used as template. The results showed higher sensitivity for Borrelia detection as compared to standard dark-field microscopy. Method specificity was confirmed by cloning and sequencing of obtained 552 base pairs long amplicons. Phylogenetic analysis of obtained sequences showed that they belong to B. lusitaniae and B. afzelii genospecies. This is the first report of the molecular characterization and identification of Borrelia burgdorferi sensu lato strains isolated in Serbia. Isolates A1, A2 and M1, from Ixodes ricinus, belong to Borrelia burgdorferi sensu stricto, while isolate K1 from Apodemus flavicollis is a mixture of Borrelia afzelii and B. burgdorferi s.s. |

We investigated the presence of Babesia microti and B. divergens in Ixodes ricinus ticks collected in a Lyme borreliosis endemic area of Northern Poland. We used as the PCR target the fragment from a gene encoding the nuclear small-subunit ribosomal RNA (SS-rDNA). We have made a thorough analysis of the quantitative and rate per cent occurrence Ixodes ricinus from two areas of forest in province of Szczecin, known as highly recreative and frequented of many people resort. A total of 533 specimens collected in 13.3% was detected DNA of B. microti and in 3.0% of B. divergens. |
